Polymer-free hydrogel made of lipid nanocapsules, as a local drug delivery platform

Nanoparticle-loaded hydrogels are attractive pharmaceutical drug delivery systems that combine the advantages of both hydrogel (local administration and/or sustained release) and nanoparticle (stealthiness, targeting decreased toxicity). The design nanoparticle-loaded is largely conventional, consisting dispersion nanoparticles in a natural or synthetic polymer matrix to form gel network. Novel architecture could provide terms innovation application. We focused on development lipid nanocapsule (LNC)-based without use as platform for delivery. Cytidine was modified by grafting palmitoyl chains (CytC16) new entity added during LNC phase-inversion formulation process allowing spontaneous gelation. Positioned at oil/water interface, CytC16 acts crosslinking agent between LNCs. Association LNCs three-dimensional network led formation polymer-free hydrogels. viscoelastic properties LNC-based depended concentration loading but were not affected size distribution. drug-release profiles controlled mechanical (slower release correlated with higher viscoelasticity). Finally, subcutaneous classic inflammatory reactions foreign body-reaction type due endogenous character CytC16, shown cellular viability assays. New-generation (LNC-based hydrogels) show promise implants applications. Once completed, no remains injection site, minimizing additional toxicity persistence polymeric implants. Sustained can be tailor-made, depending therapeutic strategy chosen.
